Esta comunicação resulta de um projeto de dissertação no campo da Ciência da Informação, área da Gestão da Informação (GI), tendo como tema central a criação de um repositório seguro no âmbito da atuação de um provedor de serviços de certificação digital: a MULTICERT, S.A.O principal objetivo fixado visou contribuir para a estruturação do repositório digital da Multicert desenvolvendo um detalhado diagnóstico organizacional e informacional e consequente elaboração e aplicação de um Documento de Especificação de Requisitos.Sendo a Multicert um provedor de serviços de certificação eletrónica, tem uma grande preocupação com a segurança da informação e pretende que esta se mantenha autêntica, integra, fidedigna, acessível e inteligível no longo prazo. In 1933, the dictatorial regime imposed in Portugal after the coup of May 1926 lost its military status. The government, since 1932 headed by Oliveira Salazar, wrote a new constitution, which was published in April 1933, and in the following months prepared a set of legislative orders that became the fundamental decrees of the new corporative state, the Estado Novo (New State). The drafts of those decrees were published in the newspaper Diário de Notícias, to allow the several social forces to react and propose changes to the legislation before it became official. One of those decrees established the principles of the construction of affordable houses by corporative or public institutions, with the support of the State. The Affordable Houses Programme was, from the start of the Estado Novo (New State), seen by the government as a central initiative in the framing of a new social order, and built over sixteen thousand houses across the country until the end of the regime in 1974. In natural environments fish populations are exposed to many potential xenoestrogens, whereby understanding the impacts of mixtures continue to be of great interest. The main objective of this study was, therefore, to understand whether and how an environmentally relevant mixture of xenoestrogens found in the Douro River estuary can disrupt the normal gametogenesis in fish. For this purpose, adult zebrafish of both sexes were exposed for 21 days to an environmental mixture (MIX) of 11 xenoestrogens from diverse sources. A 100 ng/L ethinylestradiol (EE2) positive control was added. A quantitative (stereological) analysis with systematic sampling was made in the gonads, and using light microscopy both the relative and the absolute volumes of the gametogenic stages were estimated. Data point that the EE2 stimulus induced changes in structural compartments; with decreasing trends for the advanced maturation stages both in males and females. There was also a trend for a greater amount of interstitial tissue in males. Along with an interstitial fibrosis increase detected, the presence of a proteinaceous fluid was observed in both sexes and experimental groups (EE2 and MIX). The effects of temperature on Pagellus bogaraveo muscle cellularity were investigated, by morphometry, throughout the endogenous feeding stage, testing two rearing temperatures: 14 and 18 degrees C. The following parameters were estimated in transversal body sections at post-opercular and post-anal body levels: the total cross-sectional muscle area, the total number of fibres and the mean cross-sectional fibre area. At hatching, no significant influence of the temperature was observed on the morphometric parameters measured in the white muscle. At mouth opening, an increase in the number of post-opercular white fibres was promoted by the highest temperature. During embryonic development, the red muscle fibre number in the post-anal part of the larvae increased with higher temperature, but it appears that the difference was no longer present at mouth opening. An increase in the fibre area and in the total cross-sectional area of red muscle at the post-anal level was promoted by a 4 degrees C increase in the temperature during the vitelline phase. In conclusion, the axial musculature of blackspot seabream embryos/larvae reacted differently to temperature influence according to the body location, strongly supporting the need to look at and account for different body locations when evaluating muscle cellularity in fish...

One possible approach to overcome solubility complications and enhance the biological activity of drugs is their incorporation into drug delivery systems. Within this scope, several nanosphere and nanocapsule formulations of a new inhibitor of p53-MDM2 interaction (xanthone 1) were developed and their physicochemical properties analyzed. Through the investigation of the effect of several empty nanopartides on the growth of MCF-7 cells, it was possible to observe that four out of five formulations were cytotoxic and that some correlations between the toxic potential of these polymeric nanoparticles and their properties/composition could be extrapolated. One empty formulation of nanocapsules developed by emulsification/solvent evaporation and containing PLGA, PVA and Mygliol (R) 812 was found to be noncytotoxic to this cell line. The corresponding compound 1-loaded nanocapsules showed an incorporation efficiency of 77% and revealed to be more potent than the free drug against cell growth inhibition, which may be related to the enhancement in its intracellular delivery. Previous studies have suggested that androgens may play an integral role in early oocyte development in fish. This study evaluated the effects of androgens (11-ketotestosterone: 11-KT and testosterone: T) on gene expression patterns and growth of cod previtellogenic oocytes. cDNA libraries of androgen-responsive genes were generated using suppressive subtractive hybridization (SSH) of clones containing differentially expressed genes in oocytes separately exposed to different concentrations of 11-KT and T, in addition to a solvent control. Secondly, a targeted microarray was developed based on differentially expressed genes. In the experimental setup, tissue was cultured in vitro with different concentrations of 11-KT and T (0, 10 and 100 mu M). The array analyses showed 0.5-3.5-fold significant alterations in transcript levels for a number of genes. Real-time PCR and in -situ hybridization were also used to analyze the changes in expression for selected genes. Quantitative histological analyses showed a consistent stereological validation of oocyte growth and development after exposure to androgens. The present study reveals novel roles of androgens on the development of previtellogenic oocytes, suggesting androgen control of early oocyte growth in cod. A 12-week feeding trial was conducted to evaluate the effects of fish oil replacement by soybean oil, on lipid distribution and liver histology of two commercially important finfish species: rainbow trout (Oncorhynchus mykiss) and European sea bass (Dicentrarchus labrax). Sea bass (16.2 +/- 0.5 g; mean +/- SD) and rainbow trout (52.1 +/- 0.5 g) juveniles were fed one of three isonitrogenous (500 g kg(-1) CP) and isoenergetic (19 kJ g(-1)) diets, containing 0% (control, diet A), 25% (diet B) and 50% (diet C) soybean oil. At the end of the experiment, lipid deposition was evaluated in muscle, liver and viscera. Cholesterol and triglycerides levels were also determined in plasma. Tissue total, neutral and polar lipid composition (g kg(-1) total lipids) showed no significant differences within species, regardless the dietary treatment. The same trend was observed for plasma parameters (P > 0.05). Viscera were the preferential tissue of lipid deposition, with 252-276 and 469-513 g kg(-1) total lipid content in trout and sea bass, respectively. Dietary fish oil replacement had no effect on either hepatic lipid droplets accumulation or degree and pattern of vacuolization in the observed liver sections. These data suggest that both sea bass and trout can be fed diets containing up to 50% soybean oil without adverse effects on tissue lipid composition or liver histology.

A banalização do acesso às tecnologias de informação e comunicação exige mudanças nas estratégias de ensino-aprendizagem, sob pena de inviabilizar os desígnios da educação. Enquadrada na perspetiva do conetivismo de Siemens (2005), este exercício centra-se no espaço de formação do 1º ciclo em Geografia da Universidade do Porto e a metodologia passa pela análise comparativa da resposta dos estudantes à disponibilização de recursos e estratégias didáticas em sessões presenciais (face to face) e no Facebook. Concluiu-se que a utilização desta rede constituiu um reforço no sucesso dos estudantes.

The aim of this work was to gain insights into the mechanism of muscle differentiation and growth in Pagellus bogaraveo, by studying Muscle fibre phenotypes identified by immunohistochemistry. At hatching, several layers of deep fast-white fibres were covered by a superficial fibre monolayer. At 5 days, slow-red fibres appeared near the lateral line nerve. At 40 days, the intermediate-pink muscle became visible, and in the slow-red and fast-white muscle layers transitions from larval myosin isoforms to the isoforms typical of adult muscle occurred. Between 70 and 100 days, small fibres with a distinct ATPase profile appeared throughout the fast-white muscle, marking the onset of "mosaic" hyperplasia. The myosin of the original superficial monolayer fibres underwent two myosin transformations, before being slowly replaced by an adult slow-red isoform. In juveniles and adults, the slow-red muscle layer Could be resolved into two distinct types. The analysis of fibre phenotypes indicated that post-larval muscle growth occurred by two distinct stages of hyperplasia. This study offers a basis for further comparative and experimental studies with this economically relevant species, namely for identifying factors influencing its muscle growth dynamics and disclosing underlying mechanisms.
